Abstract

±³Á¤Àû Ä¡¾ÆÀ̵¿°ú ¹ÐÁ¢ÇÑ °ü·ÃÀÌ ÀÖ´Â Ä¡ÁÖÀδ뼼Æ÷¿¡¼­, »êÈ­Áú¼Ò°¡ ¼¼Æ÷ÀÇ Áõ½Ä°ú ºÐÈ­¿¡ ¹ÌÄ¡´Â ¿µÇâÀ» ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù. Àΰ£ Ä¡ÁÖÀδ뼼Æ÷¸¦ ºÐÁÖÇÑ ÈÄ¿¡ »êÈ­Áú¼ÒÀÇ donorÀÎ SNPÀÇ ³óµµ¿¡ µû¶ó ½ÇÇ豺À» ±¸ºÐÇÏ°í, ¼¼Æ÷ È°¼º ¿°±â¼º ÀλêºÐÇØÈ¿¼Ò È°¼º, Western blot ºÐ¼®À» ÅëÇÑ osteonectin°ú bone sialoproteinÀÇ ¹ßÇö Á¤µµ¸¦ ÃøÁ¤ÇÏ¿´´Ù. 0.2 mM ÀÌÇÏÀÇ Àú³óµµ SNP¸¦ ó¸®ÇÑ ½ÇÇ豺¿¡¼­ ´ëÁ¶±º°ú ºñ±³ÇÏ¿© ¼¼Æ÷ È°¼º, ¿°±â¼º ÀλêºÐÇØ È¿¼ÒÀÇ È°¼º, ±×¸®°í osteonectin°ú bone sialoproteinÀÇ ¹ßÇöÀÌ À¯ÀÇÇÏ°Ô Áõ°¡ÇÏ¿´´Ù. ±×·¯³ª 1 mM ÀÌ»óÀÇ °í³óµµ SNP¸¦ ó¸®ÇÑ ½ÇÇ豺¿¡¼­´Â ¿ÀÈ÷·Á °¨¼ÒÇÏ¿´´Ù. »êÈ­Áú¼Ò´Â Àΰ£ Ä¡ÁÖÀδ뼼Æ÷¿¡¼­ Àú³óµµ´Â ¼¼Æ÷ÀÇ Áõ½Ä°ú ºÐÈ­¿¡ ÃËÁø È¿°ú¸¦, °í³óµµ´Â ¾ïÁ¦ È¿°ú¸¦ º¸ÀÌ´Â biphasic effect¸¦ °®´Â´Ù.

This study evaluated the effects of nitric oxide (NO) on the proliferation and differentiation of human periodontal ligament cells involved in orthodontic tooth movement.

Methods:A range of concentrations of sodium nitroprusside (SNP), a NO donor, were administered to samples of human periodontal ligament cells, followed by measurement of cell viability, alkaline phosphatase (ALP) activity, and expression of osteonectin and bone sialoprotein.

Results:Cell viability, ALP activity and the expression of osteonectin and bone sialoprotein were increased in human periodontal ligament cells treated with SNP concentrations of < 0.2 mM compared with controls, but were decreased with SNP concentrations of > 1.0 mM.

Conclusion:NO has a biphasic effect on proliferation and differentiation in human periodontal ligament cells, with a stimulatory effect at low concentrations and an inhibitory effect at high concentrations
